Abstract
Point-of-care (POC) diagnostic system involving the nucleic acid amplification test (NAAT) has gained massive popularity because of its sensitivity and specificity. Very few of these tests can deal with crude biological samples. Therefore, most of these miniaturized POC devices often rely on expensive dedicated lab equipment for sample preparation, which involves isolating and purifying nucleic acids from the complex samples, such as blood, sputum, urine, or organic tissues. To overcome this limitation, we demonstrate a POC-compatible paper-based device capable of electrochemically lysis of bacterial (M. Smegmatis) cells along with purification and concentration of genomic DNA (gDNA) from viscous mock-sputum using isotachophoresis, within 20 min. This is the first demonstration of sputum DNA preparation using paper-based ITP. © 2022 MicroTAS 2022 - 26th International Conference on Miniaturized Systems for Chemistry and Life Sciences.
