Abstract

The single-stranded RNA of the RNA viruses replicates inside the host through a mechanism in which the complementary strands are synthesized soon after infection. These minus strands assoc. with plus (viral) strands to form the double-stranded replicative intermediates which synthesize viral RNA by an asym. semiconservative mechanism. Enzyme prepns. from infected cells were used to study the replication of RNA viruses in vitro. These enzyme prepns. synthesize RNAs which are identical to primer RNA in all respects including infectivity. The reaction mixt. analyzed at varying time intervals revealed the presence of Hoffschneider structures (double-stranded) and Franklin structures (multiple-stranded contg. growing plus strands). The time course of disappearance of primer RNA and appearance of product RNA establishes the temporal sequence of RNA synthesis. The replication of viral RNA involves polymn. of the neg. strand in the 5' to the 3' direction. The synthesis of plus strands also follows the same polarity. Certain regulatory mechanisms exist which control the synthesis of the phage RNA and proteins. The coat protein inhibits the synthesis of the replicase.
