Abstract

In this paper, we demonstrate a very simple and cost effective method to fabricate three dimensional top and bottom electrodes for the application of leukocytes analysis using impedance based cytometry. The developed microfluidic impedance cytometry has compactness and no requirement of optical interface. The off-chip sample preparation was carried out to analyze the leukocytes cell differential count. Human blood was treated with saponin/formic acid to lyse erythrocytes and subsequently quench was added to halt the lysis of leukocytes. A detailed fabrication process of microelectrodes and microfluidic chip are presented in this paper. The Microfluidic Impedance Cytometry (MIC) consists of a microfluidic chip with micro patterned electrodes that measure the impedance of each cell at two frequencies and sub-population of leukocyte count was performed after recording the data from lock-in amplifier.
