This study presents the first transport-reaction model of a nucleic acid lateral flow assay (NALFA). We prove that in NALFA, in addition to the hook effect, the presence of unreacted primers in the polymerase chain reaction (PCR) product is also responsible for the reduction in test line intensity; we demonstrate this both experimentally and using the transport-reaction model. These results elucidate that the performance of a NALFA is tightly coupled to the design and conduct of the PCR reaction. © 2021 MicroTAS 2021 - 25th International Conference on Miniaturized Systems for Chemistry and Life Sciences.
