Abstract

In an in vitro system using the minces of placental villi from first trimester human pregnancy (6-10 wk), addition of low concentration of synthetic gonadotropin-releasing hormone (GnRH) [33515-09-2] or its analogs (D-Phe6-GnRH [57521-78-5] and Buserelin [57982-77-1]) $(1 \times 10^{-8}-1 \times 10^{-6}M)$ increased the quantity of human chorionic gonadotropin (hCG) [9002-61-3] in the medium, whereas addition of high concentrations of GnRH had an inhibitory effect. Of the analogs tested, Buserelin was highly effective in exerting an inhibitory effect. A significant increase in 35S-methionine incorporation into immunoprecipitable hCG was noticed in the presence of GnRH. Evidently, GnRH stimulates both synthesis and secretion of hCG by first trimester human placenta.
