The two key enzymes of triacylglycerol biosynthesis, acyl CoA synthetase and sn-glycerol-3-phosphate acyltransferase, were characterized in the developing groundnut seed. Both enzymes were found to be localized mainly in the microsomal fraction, with some activity in the mitochondrial fraction. The fat fraction, contg. the oleosomes, did not show either enzyme activity. Both enzymes showed a sharp peak of specific activity at 30 days after fertilization and this pattern closely paralleled the lipid accumulation profile in the developing seeds. The results suggest that the two enzymes are tightly regulated and induced during the period of active lipid accumulation.
