Significant similarities are now known to exist between bacterial cell division and the division of plastids in higher plants. This is primarily due to the recently proven requirement of homologues of the bacterial cell division gene ftsZ, for plastid division. This report describes the generation of three partial FtsZ cDNAs from cucumber by reverse-transcriptase mediated- polymerase chain reaction (RT-PCR) and the subsequent monitoring of the transcript levels of one ftsZ gene by high sensitivity quantitative RT-PCR, in etiolated, excised cucumber cotyledon tissue in response to the exposure to light and the exogenous application of the phytohormones, cytokinin and auxin. The results obtained clearly showed an increase in the transcript levels of the ftsZ gene examined in response to exposure to light and treatment with cytokinin, unlike in the case of auxin treatment, where no significant change was observed. These results are further discussed in terms of the possible involvement of the ftsZ gene in light- and cytokinin- induced plastid division.
